Título: Estudo da atividade antimicrobiana e cicatrizante de feridas em ratos utilizando o extrato etanólico de folhas de Tabebuia aurea (Ipê Amarelo)

Resumo: A pesquisa objetivou avaliar o potencial antimicrobiano e cicatrizante de feridas do extrato etanólico de folhas de Tabebuia aurea. O estímulo governamental às pesquisas científicas que busquem novas opções de tratamento à base de plantas medicinais, a necessidade de comprovação de eficácia e segurança terapêutica para a utilização do material vegetal pela população e que plantas do gênero Tabebuia, que possuem atividades biológicas cientificamente comprovadas e ademais, estão presentes em grande número em solo brasileiro justificam este estudo. Para a concretização dos objetivos propostos, foi realizado o preparo do extrato etanólico de folhas de Tabebuia aurea (EETA), em seguida, realizou-se seu fracionamento obtendo as frações hexânica, clorofórmica, acetato de etila e metanólica. Foram realizados testes de prospecção fitoquímica, avaliação da viabilidade celular in vitro pelo método de Metiltetrazólio, determinação da concentração inibitória mínima (CIM) para bactérias e fungos e avaliação da atividade cicatrizante de feridas cutâneas em ratos a partir da aplicação tópica do EETA a 5%. A prospecção fitoquímica foi positiva para alguns metabólitos secundários da classe dos compostos fenólicos e esteroides. O teste de citotoxidade em células da linhagem J774.A1 demonstrou citotoxidade nas concentrações de 500, 100 e 10 µg/mL e viabilidade celular na concentração de 1 µg/mL. Na avaliação da CIM, o EETA não inibiu o crescimento dos microrganismos testados, muito embora as frações hexânica e clorofórmica do EETA demonstraram atividade na concentração de 1000 µg/mL para a bactéria Escherichia coli. Nenhuma das frações inibiu o fungo Candida albicans. No estudo da atividade cicatrizante de feridas na avaliação macroscópica das feridas, o grupo EETA apresentou menor rubor perilesional e inflamação no processo inicial da cicatrização e formação de crosta espessa aderida à ferida, o que interferiu na retração das bordas apresentando percentual de contração negativo até o 11º dia de tratamento (-35,40%, -8,85% e - 1,18%) e positivo no 14º (22,71%). Os grupos controles positivo e negativo demonstraram semelhança na evolução da cicatrização, apresentando percentual de contração de 85,54% e 80,23%, respectivamente, no 14º dia. A análise histopatológica das feridas não demonstrou significância estatística na cicatrização entre os grupos no 3º e 7º dias, e significância para o grupo EETA no 14º dia, mostrando reepitelização inferior aos grupos positivo e negativo relacionado à formação de crosta. A avaliação macroscópica e histopatológica do fígado e dos rins não apresentou sinais de toxicidade assim como não houve alteração na avaliação bioquímica, indicando que o EETA utilizado topicamente por 14 dias não promove danos no modelo animal utilizado. Conclui-se que o EETA não possui atividade antimicrobiana e as frações hexânica e clorofórmica possuem fraca atividade para a bactéria E. coli; demonstrou citotoxicidade in vitro em concentração a partir de 10 µg/mL e ausência de toxicidade in vivo; sua atividade cicatrizante não foi superior aos controles avaliados neste estudo. Sugere-se investir em estudos que avaliem a atividade anti-inflamatória do EETA.
Abstract: The research aimed to evaluate the antimicrobial potential and healing of wounds of the ethanol extract of leaves of Tabebuia aurea. The government stimulus to scientific research to seek new treatment options based on medicinal plants; the need for proof of efficacy and safety for the use of plant material by the population; and Tabebuia genus plants have scientifically proven biological activities and in addition, are present in large numbers on Brazilian soil justify this study. To achieve the proposed objectives was carried out the preparation of the ethanol extract of leaves of Tabebuia aurea (EETA), and then held its fractionation, obtaining the fractions hexane, chloroform, ethyl acetate and methanol. Phytochemical tests were conducted assessing cell viability in vitro by Methyl tetrazolium method of determining the minimum inhibitory concentration (MIC) for bacteria and fungi and evaluation of the healing activity of skin wounds in rats from topical application of EETA 5%. The phytochemical screening was positive for some secondary metabolites of the class of phenolic compounds and steroids. The cytotoxicity test J774.A1 lineage cells showed cytotoxicity at concentrations of 500, 100 and 10 µg/mL and cell viability at the concentration of 1 µg/mL. In assessing the MIC, the EETA did not inhibit the growth of microorganisms tested, although the hexane fractions and chloroform the EETA showed activity at 1000 µg/mL for Escherichia coli bacteria. None of the fractions inhibited the fungus Candida albicans. In the study of healing activity healing, the macroscopic assessment of wounds, the EETA group had lower redness close to the lesion and inflammation in the initial process of wound healing and formation of a thick crust adhering to the wound, which interfere with the retraction of the edges, with the percentage of negative contraction until the 11th day of treatment (-35.40%, -8.85% and -1.18%) and positive in 14th (22.71%). The positive and negative control groups showed similarity in the evolution of healing, with the percentage of contraction of 85.54% and 80.23%, respectively, on the 14th day. The histopathology of the wounds showed no statistical significance in healing between groups at 3 and 7 days, and significance to the EETA group on the 14th day, showing reepitelization below the positive and negative groups, related to the formation of crust. Macroscopic and histopathological evaluation of liver and kidney, showed no signs of toxicity and no change in biochemical evaluation, indicating that the EETA used topically for 14 days does not promote damage to the animal model used. Concludes that the EETA not have antimicrobial activity and hexane fractions and chloroform have weak activity to the bacterium E. coli; demonstrated cytotoxicity in vitro at concentrations from 10 µg/mL and no in vivo toxicity; his healing activity was not superior to controls in this study. It is suggested invest in studies evaluating the anti-inflammatory activity EETA.
